Pregabalin

Pregabalin is a neuropathic pain reliever and anticonvulsant commonly used to treat conditions like neuropathic pain, seizures, generalized anxiety disorder (GAD), and fibromyalgia. It works by affecting the way the nervous system transmits pain signals and reducing abnormal brain activity.

  • Recommended Usage
Indications:
  • Neuropathic pain: For conditions like diabetic neuropathy and postherpetic neuralgia.
  • Seizures: As an adjunctive treatment for partial seizures in adults and children (aged 1 and older).
  • Generalized Anxiety Disorder (GAD): Pregabalin can help reduce anxiety symptoms.
  • Fibromyalgia: To manage chronic pain and muscle tenderness associated with fibromyalgia.
  • Restless Legs Syndrome (RLS): Occasionally used to treat RLS when symptoms are severe.
Dosage and Administration:
  • Neuropathic pain: Typically started at 150 mg/day, divided into 2-3 doses, and may increase to 300-600 mg/day based on response and tolerability.
  • Seizures: Starting dose is usually 150 mg/day, which can be increased to 600 mg/day in divided doses.
  • Anxiety: Typically 150 mg/day, and can be increased to 600 mg/day.
  • Fibromyalgia: Usually starts at 75 mg twice daily, with the potential to increase to 150 mg twice daily.

Pregabalin is taken orally, with or without food, and capsules should be swallowed whole.

Precautions:
  • Kidney function: Pregabalin is excreted via the kidneys, so dose adjustments are necessary for individuals with renal impairment.
  • CNS effects: It can cause drowsiness, dizziness, and cognitive impairment, so caution is needed for activities requiring alertness.
  • Withdrawal: Abrupt discontinuation can lead to withdrawal symptoms, including insomnia, headache, and nausea.
  • Pregnancy and breastfeeding: Pregabalin is Category C during pregnancy (risk cannot be ruled out) and should be used during breastfeeding only if the benefits outweigh the risks.
Contraindications:
  • Hypersensitivity to pregabalin or its components.
  • Severe renal impairment: Requires dose adjustments in kidney dysfunction, and may be contraindicated in cases of severe renal disease.
